Principle of the Assay
The HiLA™ assay is based on a bead-based sandwich immunoassay using chemiluminescent detection. The target cytokine is simultaneously captured by an F-conjugated antibody and a biotin-conjugated antibody, forming a sandwich complex.
This immune complex binds to acceptor beads and is subsequently incubated with streptavidin-coated donor beads. When the beads are brought within close proximity (<200 nm), excitation at 680 nm generates singlet oxygen, resulting in light emission at 615 nm. The emitted signal is directly proportional to the cytokine concentration.
